Maybe you get a PHP error inside the AJAX request?

I attached a screenshot on how to get further information with the Firefox plugin "Firebug".

Please open the AJAX request there and post the information that was responded by the server. What we want to know is if there is a request where the response begins with a PHP error.

okay
I think the error was due to the directory protection.
It is gone now after deleting the dir protection.

The problem is still there with Firefox 5.0.1 but doesn't occur all the time here.

Same problem here with FF 5.0.1 and TYPO3 4.5.4.
But not with all backend users.

A customer reported that he had this problem with all browsers. After disabling his proxy www-proxy.t-online.de ist worked again.
